<p>Target of replication: <em>A priori</em> experimental protocols, materials, and confirmatory analysis plans were peer reviewed, <a href="https://osf.io/j9bnk/" rel="nofollow">pre-registered</a>, and published by <a href="https://elifesciences.org/articles/13620" rel="nofollow"><em>eLife</em></a>.</p> <p>This component includes all primary and QC data for the replication of <a href="https://www.nature.com/articles/nature11005" rel="nofollow">Garnett et al. 2012</a>. This experiment analyzed Olaparib sensitivity in cells transformed with the EWS-FLI1 rearrangement.</p> <h3>Contents</h3> <p><a href="https://osf.io/4ccz9/wiki/home/" rel="nofollow">Figures component</a> contains a visualization of the results. These figures are the same as can be found in the Replication Study and include: - <strong>Figure 2. Olaparib sensitivity in cells transformed with the EWS-FLI1 rearrangement.</strong>, which displays the results of the cell viability assay. - <strong>Figure 2 - figure supplement 1. Population doubling time of cells and confirmation of EWS-FLI1 rearrangement</strong>, whicih displays the relative viability of untreated and vehicle control treated cells used to determine the population doubling times and the relative expression levels of EWS-FLI1 and Actb were determined by qRT-PCR for each cell line. </p> <p><a href="https://osf.io/5j89g/wiki/home/" rel="nofollow">Data Component</a>, contains data generated from the experimental work. Csv files containing the data used in the analysis of protocol 2, which assesses Olaparib sensitivity in cells transformed with the EWS-FLI1 rearrangement, can be found <a href="https://osf.io/k95ag/" rel="nofollow">here</a> for the qPCR data used in analysis and <a href="https://osf.io/dqy7q/" rel="nofollow">here</a> for the viability data used in analysis.</p> <p><a href="https://osf.io/a69tk/wiki/home/" rel="nofollow">Statistical Analysis Component</a>, contains the R scripts used for the analysis.</p> <p><a href="https://osf.io/egzwh/wiki/home/" rel="nofollow">Methods Component</a> includes all methods, information about equipment used, and lab notes necessary for completing this protocol.</p> <p><a href="https://osf.io/pfh3x/wiki/home/" rel="nofollow">Cell line quality Control data</a> includes results confirming the cell line was free of mycoplasma contamination as well as STR DNA profiling.</p>
