Description: In the current analysis plan, we propose to explore associations of pregnancy metabolic profiles quantified by Nuclear Magnetic Resonance (NMR) and the risk of congenital heart diseases (CHDs) in the offspring. This could help us identify biological pathways associated with CHDs and improve our understanding of the underlying pathogenesis of the condition(s). We have access to pregnancy NMR data from two large European birth cohorts from different regions which will allow us to explore replication. We will also look for genetic evidence in multiple independent cohorts to support observational results. This involves building genetic risk scores for metabolic traits using data from the largest metabolomics genome-wide association study (GWAS) and exploring associations between the risk score and CHDs.
